İnsan mikrobiyomunun karanlık kutusu: Virobiyota ve ViromGulendam Bozdayi, Işıl FidanGazi Üniversitesi Tıp Fakültesi, Tıbbi Mikrobiyoloji Anabilim Dalı, Tıbbi Viroloji Bilim Dalıİnsan mikrobiyomunun viral komponenti ‘virobiyota’ olarak tanımlanır. Virobiyota, insanda bulunan virusların toplamını ifade eder. Virobiyotadaki tüm genler ‘virom’ olarak tanımlanır. İnsan viromu, ökaryotik hücreleri enfekte eden viruslar, prokaryotik hücreleri enfekte eden bakteriyofajlar ve endojen retroviruslar gibi insan genomunda bulunan virusten türemiş genetik elementlerden oluşur. ‘High-throughput sequencing' gibi yeni sekanslama teknolojilerinin geliştirilmesi, insan genomunun analizine olanak sağlamıştır. Yeni nesil sekanslama yöntemi ile pek çok yeni virus bulunmuştur. Son yıllarda, hastalıklarda virom değişikliklerinin gözlenmesi, insan virom çalışmalarında bir artışa neden olmuştur. İnsan viromundaki değişiklikler; enfeksiyon, inflamatuvar hastalıklar, kanser ve otoimmünite ile ilişkili olabilir. Viromun insan sağlığını ve hastalığını nasıl etkilediğinin anlaşılması, virom üyelerini hedefleyen potansiyel terapötik yaklaşımların geliştirilmesini sağlayabilir. The virobiota is the collection of all viruses that are found in or on humans. The set of all the genes of the virobiota is referred to as the ‘virome’. The human virome is constituted of viruses that infect eukaryotic cells, bacteriophages, that infect prokaryotic cells, and virus-derived genetic elements in human genome, such as endogenous retroviruses. The development of new sequencing technologies, such as high-throughput sequencing techniques allowed the analysis of the human virome. Many new viruses have been discovered in recent years using new generation sequencing technology. In recent years, there has been an increase in the studies of the human virome as changes in virome have been observed in diseases. The alterations in the human virome may be associated with infectious, inflammatory diseases, cancer and autoimmunity. The understanding of how the virome affects human health and disease can provide the development of potential therapeutic approaches that target members of the virome.
