GİRİŞ ve AMAÇ: Bu çalışmada vajinal kültürlerden izole edilen Lactobacillus sp. metabolitlerinin Pseudomonas aeruginosa biyofilmleri üzerindeki antibiyofilm aktivitelerinin araştırılması amaçlanmıştır
YÖNTEM ve GEREÇLER: Çalışmada önceden 16S rRNA dizi analizi ile identifikasyonları yapılmış olan 20 adet Lactobacillus sp. izolatı kullanılmıştır. İzolatların metabolitlerinin antibiyofilm aktiviteleri P. aeruginosa üzerine mikroplak temelli antibiyofilm yöntemi kullanılarak belirlenmiştir. Negatif kontrol olarak steril De Man-Rogosa Sharpe Broth besiyeri kullanılmıştır.
BULGULAR: Test edilen metabolitlerin tamamı P. aeruginosa biyofilmleri üzerinde istatiksel olarak anlamlı antibiyofilm aktivite göstermiştir. Metabolitler arasında fark olmakla birlikte, biyofilm hücre sayısında en az 4 logaritma (kob/ml) azalış saptanmıştır. En iyi antibiyofilm aktivite L. gasseri ve L. jensenii metabolitleri için saptanmış olup, bunlar P. aeruginosa biyofilm hücrelerinin tamamını yok edebilmiştir.
TARTIŞMA ve SONUÇ: Patojen bakterilerin planktonik formlarının tedavisinde yaygın olarak kullanılan antibiyotikler, biyofilm içindeki bakterilerin eradikasyonunda çoğu zaman yetersiz kalmaktadır. Vajinal Lactobacillus sp. metabolitlerinin göstermiş olduğu iyi antibiyofilm aktivite, bu metabolitlerin biyofilm ilişkili enfeksiyonlar ile mücadelede alternatif ajanlar olarak kullanılabileceklerini düşündürmektedir.

INTRODUCTION: The aim of this study was to investigate the antibiofilm activities of metabolites produced by vaginal Lactobacillus sp. against Pseudomonas aeruginosa biofilms. METHODS: In this study, 20 Lactobacillus sp. isolates which were previously identified in terms of species level by analyzing the 16S rRNA gene sequence were used. Antibiofilm activity of the metabolites of the isolates was determined by using microplate-based antibiofilm method on P. aeruginosa. De Man-Rogosa Sharpe Broth was used as negative control. RESULTS: All tested metabolites showed statistically significant antibiofilm activity on P. aeruginosa biofilms. Although there were differences between the metabolites, the number of biofilm cells decreased at least 4 logarithms (cfu/ml). The best antibiofilm activity was determined for L. gasseri and L. jensenii metabolites destroyed all P. aeruginosa biofilm cells. DISCUSSION AND CONCLUSION: Antibiotics commonly used in the treatment of planktonic forms of pathogenic bacteria are often insufficient for the eradication of bacteria in the biofilm. Vaginal Lactobacillus sp. metabolites have shown good antibiofilm activity, suggesting that these metabolites can be used as alternative agents to help combat with biofilm-associated infections.
