GİRİŞ ve AMAÇ: Bu çalışmada öncelikle Epidermal büyüme faktör’ün (EGF) osteosarkoma hücre proliferasyonu ve Bcl-2 ekspresyonu üzerine etkileri araştırılmıştır. YÖNTEM ve GEREÇLER: Bu çalışmada ilk olarak EGF’nin hücre proliferasyonu üzerine etkisinin belirlenmesi için MTT testi uygulanmıştır. Saos-2 hücre hattına 20 ng/µl EGF eklenip, farklı inkübasyon aralıklarından sonra, hücre proliferasyonu üzerine etkileri belirlendi. Bir sonraki deneysel aşama; EGF osteosarkom hücre hattı üzerine 20ng/ml eklendi, 1, 3, 24, 48 ve 72 saat sonunda antiapopitotik Bcl-2’nin ekspresyonu değerlendirildi. BULGULAR: Osteosarkom hücre hattında 20 ng/µl EGF uygulaması ile 3, 24 ve 72 saatlerde hücre proliferasyonunda istatistiksel olarak anlamlı artış gözlendi. EGF 20ng/ml dozda 1 ve 24 saat uygulama sonunda Bcl-2 ekspresyonunu arttırdığı gözlendi. TARTIŞMA ve SONUÇ: EGF; 3, 24 ve 72 saatlerde hücre proliferasyonunda istatistiksel olarak anlamlı artışa neden olmuştur. Ayrıca EGF osteosarkom hücre hattı üzerinede, 1, 3, 24, 48 ve 72 saat sonunda antiapopitotik Bcl-2’nin ekspresyonu değerlendirilmiştir. EGF 20ng/ml dozda 1. ve 24. saat uygulama sonunda Bcl-2 ekspresyonunu artmıştır.

INTRODUCTION: In this study, effects of epidermal growth factor (EGF) on osteosarcoma cell proliferation and Bcl-2 expression were investigated. METHODS: In this study, MTT test was first applied to determine the effect of EGF on cell proliferation. 20 ng / μl EGF was added to the Saos-2 cell line and after different incubation intervals, the effects on cell proliferation were determined. The next experimental stage; 20 ng / ml was added to the EGF osteosarcoma cell line, and the expression of antiapoptotic Bcl-2 was evaluated at 1, 3, 24, 48 and 72 hours. RESULTS: A statistically significant increase in cell proliferation was observed at 3, 24 and 72 hours with 20 ng / μL EGF administration in the osteosarcoma cell line. It was observed that EGF stimulated Bcl-2 expression at the dose of 20 ng / ml for 1 and 24 hours. DISCUSSION AND CONCLUSION: EGF; At the 3 rd, 24 th and 72nd hours, a statistically significant increase in cell proliferation was observed. In addition, the expression of antiapoptotic Bcl-2 was evaluated at the 1st, 3rd, 24th, 48th and 72th hours on the EGF osteosarcoma cell line. The expression of Bcl-2 was increased at the end of the 1st and 24th hours of application of EGF at 20 ng / ml dose.
